Will the book packs last for 1 year or 2?

It depends on how often you do DITHOR. If you do it 5 days a week, they'll last 1 year. If you do it 3 days a week, you'll cover 5 genres, so a book pack would last 2 years (plus you'd have to have a book for one more genre the 2nd year). The Preparing guide tells you to use DITHOR 3 times a week.

We're using the level 2 pack this year and we'll be through it in a year because we're doing DITHOR daily. You may have noticed that some of the genres in the packs have more than one book. This is because they are books that won't take the whole 15 days of the genre to read. Carrie has a schedule you'll get with the pack that tells you how many days per book she's reccommending. I think that's actually in the book description on the website, too.

Do you still read ,say, Biography for 15 days, if you do it for 3 or 5 days?

Yep. In the teacher guide for DITHOR, there are 15 days of plans per genre. (Plus kickoff ideas that are fun! And, 3 project options to wrap up the genre. The project is to be worked on for 5 days during "reading" time.) So, if you were using DITHOR 5 days per week, you'll spend 4 weeks total on Biography. If you're using DITHOR 3 days per week, you would spend 6 1/2 weeks on Biography.
